Overview

CAWP-1322 is a small molecule inhibitor of poly(ADP-ribose) polymerase 1 (PARP-1) currently under development by Waverley Pharma. PARP-1 is an enzyme that plays a pivotal role in the repair of single-strand DNA breaks through the base excision repair pathway. By inhibiting this enzyme, CAWP-1322 prevents the repair of DNA damage, leading to the formation of double-strand breaks. In cancer cells that already possess deficiencies in homologous recombination repair (such as those with BRCA mutations), this accumulation of DNA damage leads to synthetic lethality. CAWP-1322 is being investigated in preclinical studies for its potential therapeutic application in several oncology indications, including ovarian cancer, triple-negative breast cancer, melanoma, breast cancer, and lung cancer.
