CAWP-1477 is a small molecule inhibitor of poly(ADP-ribose) polymerase 1 (PARP-1) currently in preclinical development by Waverley Pharma. PARP-1 is a critical enzyme involved in the detection and repair of single-strand DNA breaks through the base excision repair pathway. By inhibiting PARP-1, CAWP-1477 prevents the repair of these lesions, which can then lead to the formation of double-strand breaks during DNA replication. In cancer cells with existing deficiencies in homologous recombination repair (such as those with BRCA mutations), this leads to genomic instability and cell death, a concept known as synthetic lethality. CAWP-1477 is being investigated for its potential in treating several solid tumors, including ovarian cancer, triple-negative breast cancer, melanoma, breast cancer, and lung cancer.
