Overview

CB-011 is an allogeneic, off-the-shelf anti-BCMA chimeric antigen receptor (CAR) T cell therapy developed by Caribou Biosciences for the treatment of relapsed or refractory multiple myeloma. It is manufactured from healthy donor T cells that are genome-edited using Cas12a chRDNA technology to make four specific edits: knockout of the TRAC gene to remove the native T cell receptor, insertion of a humanized anti-BCMA CAR into the TRAC locus, knockout of endogenous B2M, and insertion of a B2M–HLA-E fusion protein into the B2M locus. This immune cloaking strategy eliminates endogenous HLA class I presentation and expresses HLA-E to reduce both T cell and natural killer (NK) cell-mediated rejection by the patient’s immune system. The goal is to improve persistence and antitumor activity in patients with multiple myeloma who have relapsed after or are refractory to prior therapies. CB-011 has received FDA Fast Track and Orphan Drug designations and is currently being evaluated in a Phase 1 clinical trial (CaMMouflage study) in adults with relapsed or refractory multiple myeloma[1][2][3][5][6][8].
