Overview

CB-012 is an allogeneic, genome-edited CAR-T cell therapy developed for the treatment of relapsed or refractory acute myeloid leukemia (r/r AML). It targets C-type lectin-like molecule-1 (CLL-1, also known as CD371), which is expressed on AML tumor cells and leukemic stem cells but not on normal hematopoietic stem cells. The product uses Caribou Biosciences' Cas12a chRDNA genome-editing technology to introduce five specific edits, including PD-1 knockout for checkpoint disruption and B2M knockout with B2M–HLA-E fusion insertion for immune cloaking. These modifications are designed to enhance antitumor activity by improving persistence and reducing immune rejection of the CAR-T cells. CB-012 is currently being evaluated in the AMpLify Phase 1 clinical trial and has received FDA Fast Track designation[1][3][4][5][7].
