CB-020 is a preclinical, induced pluripotent stem cell (iPSC)-derived, allogeneic CAR-NK cell therapy candidate developed by Caribou Biosciences for the treatment of solid tumors. It is engineered using Caribou’s Cas12a chRDNA genome-editing platform to derive natural killer cells from iPSCs and to express a chimeric antigen receptor specific for receptor tyrosine kinase-like orphan receptor 1 (ROR1), a tumor-associated antigen overexpressed on various solid tumors where it promotes tumor growth, survival, and metastasis.[3][4][7] The anti-ROR1 CAR is intended to enhance NK cell specificity and antitumor function against ROR1-positive tumors, as part of a broader “armored” CAR-NK platform for solid tumors, although Caribou has since paused and then stopped development of this CAR-NK program as part of portfolio reprioritization.[3][5][8][12]
