Overview

CB1093 is a **novel synthetic analog of vitamin D3 (calcitriol)**, specifically a 20-epi-vitamin D3 analog, designed to modulate cell differentiation and inhibit proliferation in various cancer cell types, such as **acute myeloid leukemia (AML) cells** and breast cancer cells[1][3][4][6]. CB1093 induces differentiation and apoptosis in cancer cells, likely through activation of the vitamin D receptor (VDR), and exhibits minimal hypercalcemic activity compared to native calcitriol, making it a promising antineoplastic agent for preclinical studies[1][4][6]. It also induces nerve growth factor (NGF) expression, which has been shown to counteract diabetic neuropathy in animal models[3]. In breast cancer models, CB1093 enhances the antitumor activity of agents like paclitaxel and cisplatin[4]. Mechanistically, CB1093 promotes cPLA2 activation and augments apoptosis both alone and in synergy with TNF-alpha and ceramide in breast cancer cell lines[6].
