CB4211 is a novel, synthetic peptide drug under development as a first-in-class mitochondria-based therapeutic (MBT) for the treatment of nonalcoholic steatohepatitis (NASH) and obesity. It is an optimized analog of MOTS-c, a naturally occurring mitochondrial-derived peptide that plays a significant role in metabolic regulation. Discovered by CohBar scientists and based on research from the Albert Einstein College of Medicine and University of Southern California, CB4211 acts primarily by modulating insulin receptor signaling and inhibiting lipolysis to maintain metabolic homeostasis. In preclinical models and early clinical trials, it has demonstrated potential to reduce liver fat, improve biomarkers of liver injury (ALT/AST), enhance insulin sensitivity, decrease glucose levels, and trend toward weight loss. As of late 2023, it remains in Phase 1 clinical trials for both NASH and obesity[2][4][5][6].
