**CB5005** is a multifunctional synthetic peptide used primarily as a *cell-penetrating peptide* (CPP) with integrated nuclear-targeting and NF-κB-inhibitory properties. It comprises a unique membrane-permeable sequence (CB5005M) linked to a nuclear localization signal that inhibits activation of nuclear factor kappa-B (NF-κB), which plays a role in cancer progression and chemoresistance. CB5005 itself can inhibit tumor growth and displays synergy in enhancing the delivery and efficacy of chemotherapeutics (such as doxorubicin and irinotecan) when conjugated to nanoparticle drug carriers, including PEGylated liposomes. As a dual-functional synergy agent, CB5005 enables deeper tumor penetration, increased nuclear drug delivery, and direct inhibition of nuclear NF-κB function, improving antitumor activity—in vivo and in vitro—in various cancer models such as glioma and non-small cell lung carcinoma[1][3][5][7].
