Overview

CB699 is a half-life extended, trispecific multi-specific antibody developed by Crescendo Biologics. It targets three proteins—CD40 (tumor necrosis factor receptor superfamily member 5), CD137 (also known as 4-1BB, tumor necrosis factor receptor superfamily member 9), and mesothelin (MSLN). The molecule is designed to act as a dual agonist for CD40 and CD137 in a mesothelin-directed manner, conditionally activating T cells and antigen-presenting cells (APCs) only in the presence of MSLN+ tumor cells. This conditional activation aims to provide potent anti-cancer immune responses while minimizing off-tumor toxicity. Preclinical data show that CB699 can drive both CD40 and CD137 agonism depending on the cellular context, enhancing immune cell signaling axes for comprehensive anti-tumor activity[1][6][8]. The drug is currently in preclinical development for the treatment of mesothelin-positive neoplasms.
