CBA-1205 is a novel glycoengineered humanized monoclonal antibody that targets delta-like 1 homolog (DLK1), a non-canonical Notch ligand highly expressed in various malignant tumors, especially hepatocellular carcinoma. The antibody is afucosylated to enhance antibody-dependent cellular cytotoxicity (ADCC) and selectively binds to DLK1 between its epidermal growth factor-like domains 1 and 2. CBA-1205 has demonstrated potent antitumor activity in preclinical models of liver cancer and other solid tumors, both as a single agent and in combination with lenvatinib. In clinical studies, it has shown favorable safety profiles with no dose-limiting toxicities or severe adverse events reported at doses up to 30 mg/kg. The drug is being developed primarily for the treatment of advanced or recurrent solid tumors, including hepatocellular carcinoma.
