Overview

CBB-120 is a next-generation, dual-payload antibody-drug conjugate (ADC) developed by CrossBridge Bio for the treatment of TROP-2-positive solid tumors, including triple-negative breast cancer and other cancers with unmet needs. The drug leverages a proprietary, highly stable tripeptide linker technology that enables site-specific conjugation of two distinct cytotoxic payloads to an anti-TROP-2 antibody. This design aims to maximize therapeutic efficacy by delivering synergistic effects within targeted cancer cells while minimizing off-target toxicity and overcoming tumor heterogeneity and resistance mechanisms. The ADC targets TROP-2 (tumor-associated calcium signal transducer 2), a protein overexpressed in various epithelial cancers, using dual mechanisms: inhibition of serine-protein kinase ATR (ATR inhibitor) and DNA topoisomerase I (TOP1 inhibitor). CBB-120 is currently in preclinical development[1][4][5][7].
