Overview

CBD-alphaGITR is an engineered collagen-binding monoclonal antibody designed to target the Glucocorticoid-induced TNFR-related protein (GITR) specifically within the tumor microenvironment. It consists of an anti-GITR antibody conjugated to the collagen-binding domain (CBD) derived from von Willebrand Factor (vWF), which enables selective accumulation in tumors expressing high levels of Collagen type I and III, such as recurrent glioblastoma (rGBM), while sparing normal brain tissue. By agonistically targeting GITR, the drug reduces the population of regulatory T cells (Tregs) and exhausted CD8+ T cells, thereby remodeling the immunosuppressive microenvironment and enhancing anti-tumor immune responses. It has shown significant efficacy in preclinical models of rGBM, particularly when combined with PD-1 inhibitors.
