Overview

CBD-anti-CTLA4 is an engineered recombinant fusion protein designed to improve the therapeutic index of CTLA-4 checkpoint inhibition. It consists of an anti-CTLA4 monoclonal antibody fused to a collagen-binding domain (CBD) derived from the von Willebrand Factor (vWF) A3 domain. This construct is designed to target the tumor microenvironment by binding to exposed collagen in the tumor stroma after intravenous administration. By localizing the antibody specifically within tumor tissues, the drug achieves deep tissue infiltration and efficient depletion of intratumoral regulatory T cells (Tregs) while significantly reducing systemic exposure and associated toxicities, such as hepatotoxicity. Developed by researchers at Imperial College London, CBD-anti-CTLA4 has demonstrated superior anti-tumor efficacy and complete tumor regression in preclinical models of glioblastoma, melanoma, and breast cancer.
