CBD-IL-12-expressing FAP CAR-T is an engineered chimeric antigen receptor (CAR) T-cell therapy designed to target the fibrotic tumor microenvironment. It specifically targets fibroblast activation protein (FAP) expressed on cancer-associated fibroblasts (CAFs). Upon binding to FAP, the CAR-T cells utilize NFAT signaling to trigger the localized secretion of interleukin-12 (IL-12) fused to a collagen-binding domain (CBD). This dual-action approach aims to deplete CAFs, thereby reducing the physical barrier to immune cell infiltration, while simultaneously concentrating IL-12 within the tumor stroma to enhance host immune activation. The CBD fusion minimizes systemic exposure and toxicity by ensuring the cytokine remains localized to the collagen-rich tumor matrix.
