Overview

CBL-0304 is a small molecule belonging to the Curaxin family, a class of compounds originally identified for their ability to simultaneously activate p53 and inhibit NF-κB without inducing DNA damage. The primary mechanism of action involves high-affinity, non-genotoxic intercalation into DNA, which alters the physical properties of the chromatin template. This structural change leads to the sequestration of the facilitates chromatin transcription (FACT) complex on the DNA, a process termed chromatin remodeling. The loss of functional FACT activity prevents the transcription of NF-κB-dependent genes and triggers a p53 response. Developed by Cleveland BioLabs and Incuron, CBL-0304 served as a prototype for the Curaxin class, demonstrating potent anti-tumor activity in preclinical models of various cancers, including colon, breast, and renal cell carcinomas, before the clinical focus shifted to the optimized analog CBL0137.
