Drug intelligence / Profile preview
CBL-514 + tirzepatide is an investigational combination therapy being developed by Caliway Biopharmaceuticals for localized fat reduction, weight management, and the prevention of post-treatment weight regain. The combination pairs CBL-514, an injectable lipolysis agent composed of curcumin and trans-resveratrol, with tirzepatide, a dual GIP and GLP-1 receptor agonist marketed as Zepbound and Mounjaro. CBL-514 selectively induces adipocyte apoptosis by inhibiting the cell survival kinase DYRK1B, permanently reducing the number of fat cells in targeted subcutaneous and visceral depots. Tirzepatide primarily regulates appetite and energy intake. In preclinical studies, the combination demonstrated synergistic effects, including enhanced subcutaneous and visceral fat reduction, improved hepatic lipid parameters, and a significant reduction in weight regain after tirzepatide discontinuation compared to tirzepatide alone. Caliway is evaluating the combination in the Phase 2 clinical trial CBL-0201WR in overweight or obese adults.
