Overview

CBLB612 is a synthetic lipopeptide drug that acts as an agonist of Toll-like receptor 2 (TLR2) and Toll-like receptor 6 (TLR6). It is being developed primarily for the induction and mobilization of hematopoietic stem cells (HSCs), with potential applications in enhancing HSC collection for transplantation and as a protective or restorative agent against chemotherapy-induced myelosuppression. Mechanistically, CBLB612 activates pro-survival and immunoregulatory signaling pathways via specific binding to TLR2/6 on certain cell types, leading to increased production of cytokines such as granulocyte colony-stimulating factor (G-CSF), interleukins IL-6, IL-8, IL-10, and interferon-alpha. Preclinical studies have shown that it can induce greater concentrations of HSCs in peripheral blood than currently available mobilizing drugs. Clinical trials have focused on its use for neutropenia prophylaxis in breast cancer patients undergoing myelosuppressive chemotherapy[2][5][6][7][8].
