HotSpot Therapeutics is developing oral, small molecule allosteric inhibitors targeting the CARD11-BCL10-MALT1 (CBM) signalosome complex. Discovered via the proprietary Smart Allostery™ platform, these inhibitors are designed to treat KRAS G12X-driven solid tumors, including colorectal, pancreatic, and lung cancers. The CBM signalosome acts as a molecular hub regulating multiple oncogenic pathways such as NFκB, JNK, mTORC1, and MYC. HotSpot has identified a synthetic lethal relationship where KRAS G12X mutations render tumors dependent on the CBM signalosome for survival. Preclinical data indicates these inhibitors can induce apoptosis in KRAS G12 colorectal cancer cells and show efficacy as both monotherapy and in combination with KRAS inhibitors.
