Overview

Cord blood-derived natural killer (CBNK) cells are a cell therapy product consisting of **natural killer (NK) cells** isolated and expanded from umbilical cord blood. These cells can be further modified, for example with chimeric antigen receptors (CAR), to enhance their antitumor activity. CBNK cells target and kill malignant or infected cells through both direct cytotoxicity (mediated by release of perforin and granzyme, induction of apoptosis via Fas/FasL or TRAIL) and by secretion of cytokines like IFN-γ and TNF-α, which activate additional immune responses. CBNK cells display potent antitumor effects, broad applicability in cancer, and lower risk of graft-versus-host disease compared to other cell therapies. They can be used as “off-the-shelf” allogeneic therapies. They are in clinical trials for hematologic malignancies, lymphomas, leukemias, neuroblastoma, sarcomas, and some solid tumors[2][3][4][8][12][14]. CBNK cells are not a small molecule drug but a living cell therapy product, often manufactured by academic centers or biotech companies in collaboration with clinical research institutions.
