CBO-002 is a preclinical-stage nanobody-drug conjugate (NDC) being developed by CIS Biopharma AG for the treatment of brain metastases. The therapeutic consists of a nanobody (VHH antibody fragment) that specifically targets CD276 (also known as B7-H3), an immune checkpoint molecule frequently overexpressed in various solid tumors and metastatic lesions. This nanobody is conjugated to a Topoisomerase I (Topo-I) inhibitor payload. The NDC modality is designed to combine the high specificity and small size of nanobodies—which facilitates superior tissue penetration compared to traditional antibodies—with the potent cytotoxic effects of a chemotherapy payload. This approach is intended to improve the delivery of the cytotoxic agent to difficult-to-reach tumors, such as those located in the brain.
