Overview

CBO421 is a first-in-class drug-Fc conjugate (DFC) immunotherapy developed by Cidara Therapeutics for the treatment of solid tumors. It targets CD73, a cell surface enzyme (5'-nucleotidase) responsible for the production of immunosuppressive adenosine, which enables tumor immune evasion. The molecule combines features of small molecule inhibitors and monoclonal antibodies, offering potent inhibition of both soluble and membrane-bound CD73. Its mechanism includes catalytic inhibition of CD73 enzymatic activity as well as induction of receptor internalization and degradation, thereby reducing adenosine-mediated immunosuppression in the tumor microenvironment. Preclinical studies have shown robust antitumor efficacy both as monotherapy and in combination with PD-1 inhibitors, with superior potency compared to existing anti-CD73 monoclonal antibodies[1][2][3][5][6].
