CBPd-171 is a potent and selective protein degrader targeting CREB-binding protein (CBP), developed by Foghorn Therapeutics. It is designed to exploit a synthetic lethal relationship in cancers harboring mutations in EP300, a closely related acetyltransferase. By selectively degrading CBP, the drug aims to treat EP300-mutant solid tumors, including endometrial, cervical, ovarian, bladder, and colorectal cancers, as well as CBP-dependent malignancies like ER+ breast cancer. CBPd-171 is formulated as a long-acting injectable (LAI) for subcutaneous administration and has demonstrated a favorable safety profile in preclinical studies, specifically sparing megakaryocytes and avoiding significant platelet count reduction.
