Overview

CBT-1 + paclitaxel + carboplatin is a combination regimen under investigation for cancer therapy. CBT-1 is an orally administered small molecule inhibitor of P-glycoprotein (P-gp, also known as multidrug resistance protein 1, MDR1, or ABCB1), a membrane transporter responsible for multidrug resistance in cancer cells. CBT-1 is designed to block P-gp-mediated drug efflux, potentially enhancing the efficacy of chemotherapies that are P-gp substrates. Paclitaxel is a microtubule-stabilizing agent that disrupts cell division, and carboplatin is a DNA cross-linking platinum compound. This combination aims to improve responses in tumors known for developing resistance, particularly through the action of CBT-1 reversing multidrug resistance and enhancing the cytotoxicity of paclitaxel and carboplatin. Preclinical and early clinical data show that CBT-1 can potentiate the effects of taxanes and platinum agents in resistant cancer models.
