Overview

CBX-250 is a first-in-class, bispecific T-cell engager (TCE) designed to target a cathepsin G (CTSG)-derived peptide presented by human leukocyte antigen (HLA), specifically the CG1 pHLA complex. This complex is abundantly expressed on leukemic cells but not on normal cells, enabling selective targeting of cancerous myeloid cells while sparing healthy hematopoietic and immune cells. The drug leverages TCR-mimetic antibody technology to recognize intracellular cancer antigens presented on the cell surface via HLA, expanding the range of targetable tumor antigens beyond traditional surface proteins. Preclinical studies demonstrate potent and specific T cell-mediated cytotoxicity against acute myeloid leukemia (AML) cell lines with minimal off-target effects or toxicity toward normal hematopoietic stem and progenitor cells. CBX-250 is being developed by Crossbow Therapeutics in collaboration with MD Anderson Cancer Center for the treatment of AML[1][2][4][5].
