Overview

A CBX5 inhibitor is a therapeutic agent designed to block the activity of Chromobox 5 (CBX5), also known as Heterochromatin Protein 1 alpha (HP1α). CBX5 is an epigenetic reader protein that binds to methylated histone H3 lysine 9 (H3K9me2/3) to facilitate the formation and maintenance of constitutive heterochromatin, leading to gene silencing. In the context of oncology, particularly small cell lung cancer (SCLC), CBX5 has been identified as a key regulator of immune evasion. Inhibition of CBX5 is thought to activate immune-response genes and derepress endogenous retroviruses, thereby remodeling the tumor microenvironment from an "immune-cold" to an "immune-hot" state. This mechanism aims to enhance the sensitivity of tumors to immune checkpoint inhibitors, such as anti-PD-1 therapies, by increasing the visibility of tumor cells to the immune system.
