CC-122 + CC-292 + rituximab is a combination investigational regimen evaluated primarily in patients with relapsed or refractory diffuse large B-cell lymphoma (DLBCL). - **CC-122 (avadomide)** is a cereblon E3 ligase modulator (CELMoD), acting as an immunomodulatory and anti-proliferative agent. - **CC-292 (spebrutinib)** is a potent, selective, covalent Bruton tyrosine kinase (BTK) inhibitor that disrupts B-cell receptor signaling, resulting in decreased proliferation and survival of malignant B cells. - **Rituximab** is a monoclonal antibody targeting CD20, expressed on the surface of B cells, and exerts antitumor effects primarily via antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The triplet was studied in the multi-arm phase Ib CC-122-DLBCL-001 trial for safety, tolerability, pharmacokinetics, pharmacodynamics, and preliminary efficacy in heavily pretreated DLBCL patients. The combination was associated with enhanced toxicity and/or insufficient improvement in responses compared to other arms, which limited its further development.
