Overview

**CC-122 + obinutuzumab** is an experimental combination therapy for relapsed or refractory non-Hodgkin lymphoma, particularly evaluated in follicular lymphoma, marginal zone lymphoma, and diffuse large B-cell lymphoma. CC-122 (avadomide) is an oral small molecule that modulates the cullin 4 ring E3 ligase complex, leading to proteasomal degradation of transcription factors Aiolos and Ikaros, resulting in both cell-autonomous antineoplastic and immunomodulatory activity on T and NK cells. Obinutuzumab is a type 2 anti-CD20 monoclonal antibody that promotes B-cell lysis through enhanced direct cell death and antibody-dependent cellular cytotoxicity. Together, they show synergistic or additive efficacy in various lymphoma models, and this regimen is undergoing Phase 1b dose escalation and expansion studies for safety and preliminary efficacy.
