Overview

CC-122 + rituximab is an investigational **combination therapy** comprising avadomide (code name CC-122), a small molecule cereblon E3 ligase modulator (CELMoD), and rituximab, a CD20-directed monoclonal antibody. Preclinical and early clinical data indicate this combination exerts **synergistic antitumor effects**: avadomide acts by modulating the cereblon E3 ubiquitin ligase to induce tumor cell apoptosis and immunomodulation, while rituximab targets CD20 on B cells for antibody-dependent cell-mediated cytotoxicity. This combination has been studied in relapsed/refractory diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma (FL), showing tolerability and preliminary antitumor activity in these difficult-to-treat populations[3][4][5][1].
