CC-122 + CC-223 is an investigational oral combination therapy studied for relapsed or refractory diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma. CC-122 (avadomide) is a cereblon E3 ligase modulator (CELMoD) that induces degradation of the transcription factors Ikaros and Aiolos, leading to apoptosis of malignant B cells and activation of T-cells. CC-223 is a small molecule that selectively inhibits mammalian target of rapamycin (mTOR) kinase, targeting both mTORC1 and mTORC2 complexes, which are critical regulators of cell proliferation and survival. This combination was evaluated in a multi-arm phase Ib clinical study (NCT02031419), where preliminary safety and tolerability were assessed. CC-122 + CC-223 was tested as one of several combinations, but was associated with enhanced toxicity and did not advance to dose expansion, unlike the CC-122 plus rituximab arm[1][3][6][8].
