CC-90011 + nivolumab is a combination therapy that has been investigated in clinical trials for advanced cancers, particularly lung cancer. This combination pairs CC-90011, an oral LSD1 inhibitor developed by Celgene (now part of Bristol Myers Squibb), with nivolumab, an established immune checkpoint inhibitor. CC-90011 is an oral, potent, selective, reversible inhibitor of lysine-specific demethylase 1 (LSD1), an enzyme involved in regulating genes important for cancer progression and cell growth. Its reversible mechanism of action may provide an advantage over other irreversible LSD1 inhibitors. Nivolumab is a monoclonal antibody designed to trigger the body's immune system to destroy tumors by blocking the PD-1 pathway. The combination was studied in a Phase 2, multicenter, open-label, multi-cohort trial (CC-90011-ST-002) that evaluated its safety and efficacy in patients with small cell lung cancer (SCLC) or squamous non-small cell lung cancer (sqNSCLC) who had progressed after one or two lines of therapies. The study included three cohorts: Cohort A (SCLC in immune checkpoint inhibitor-naïve subjects), Cohort B (SCLC in immune checkpoint inhibitor progressor subjects), and Cohort C (sqNSCLC in immune checkpoint inhibitor progressor subjects).
