CC-96191 is a trispecific antibody, also referred to as a TriNKET, developed for the treatment of relapsed or refractory acute myeloid leukemia (R/R AML). It is designed to simultaneously bind three targets—CD33, CD16a, and NKG2D. By engaging these targets, CC-96191 activates natural killer (NK) cells in the presence of CD33-positive AML cells. The co-engagement of NKG2D and CD16a enhances NK cell avidity and activation, resulting in selective cytotoxicity against leukemia cells while sparing normal monocytes. Preclinical studies have shown that its anti-leukemia activity is strictly dependent on CD33 expression and that it does not activate T cells. Compared to bispecific antibodies targeting CD33/CD3, CC-96191 induces significantly lower levels of soluble cytokines[2][6]. The drug is being evaluated as an intravenous monotherapy in early-phase clinical trials for R/R AML[1][3][5].
