CC-98282 is an orally bioavailable Cereblon E3 Ligase Modulator (CELMoD) developed by Celgene (now Bristol Myers Squibb). It functions as a molecular glue that binds to cereblon (CRBN), a component of the CRL4-CRBN E3 ubiquitin ligase complex. This binding alters the substrate specificity of the ligase, leading to the recruitment, ubiquitination, and subsequent proteasomal degradation of specific transcription factors, primarily Ikaros (IKZF1) and Aiolos (IKZF3). These proteins are essential for the survival and proliferation of B-cell malignancies. CC-98282 was primarily investigated for the treatment of relapsed or refractory aggressive B-cell lymphomas, including diffuse large B-cell lymphoma (DLBCL). However, clinical development was terminated by the sponsor for strategic reasons in 2021, rather than due to safety or efficacy concerns.
