CC312 is a trispecific T cell engager (TriTE) antibody developed by CytoCares. It is designed to simultaneously target three immune-related antigens: CD19 (on B cells), CD3 (on T cells), and CD28 (a co-stimulatory molecule on T cells). The drug consists of three single-chain variable fragments (scFvs) in tandem, enabling it to bridge tumor B cells and T cells, activate the CD3 signaling pathway, provide co-stimulation via CD28, and induce robust memory T cell proliferation while preventing T cell anergy. This mechanism aims to enhance anti-tumor immunity against B-cell malignancies and modulate immune responses in autoimmune diseases. Its action profile is similar to a "biopharmaceutical version" of CAR-T therapy but as an off-the-shelf antibody product[2][3][4][5][6].
