Overview

CC49-Br-MMAE is an antibody-drug conjugate (ADC) composed of the murine monoclonal antibody CC49, which targets tumor-associated glycoprotein-72 (TAG72), conjugated to the highly potent synthetic antimitotic agent monomethyl auristatin E (MMAE) via a bromoacetamido (Br) linker attached to reduced hinge disulfides of CC49[3][5]. TAG72 is overexpressed in a wide range of epithelial-derived cancers, including ovarian cancer, but is absent in most normal adult tissues except secretory endometrium[3][5]. The Br linker chemistry used in CC49-Br-MMAE results in a high drug-to-antibody ratio (DAR) and enhanced stability, which improves its pharmacokinetics, tumor targeting, and in vivo efficacy compared to similar ADCs using other linkers[1][3][5]. In preclinical studies in ovarian cancer xenograft models, CC49-Br-MMAE demonstrated a significant tumor growth delay and an increase in survival, evidencing its potential as a targeted therapy for TAG72-positive solid tumors[3][5].
