Overview

CC49 fusion protein refers to a recombinant fusion protein derived from the CC49 (minretumomab) monoclonal antibody, which specifically targets TAG-72, a carbohydrate epitope overexpressed on the surface of many solid tumors[1][7]. The protein combines the tumor-targeting specificity of CC49 with additional functional domains for therapeutic purposes. There are several variants, including fusion with β-lactamase (BLA) for antibody-directed enzyme prodrug therapy (ADEPT), fusion with streptavidin for pretargeted radioimmunotherapy (RIT), and fusion with interleukin-2 (IL-2) for targeted immunotherapy[1][3][4]. In ADEPT, the CC49-scFv-BLA fusion (TAB2.5) localizes to tumors, where it activates a prodrug (e.g., GC-Mel) to a cytotoxic agent selectively at the tumor site, minimizing systemic toxicity[1]. In pretargeted RIT, the CC49-(scFv)4-streptavidin fusion binds tumor cells, followed by administration of a clearing agent and a radiolabeled biotin (e.g., 90Y/111In-DOTA-biotin), enabling precise delivery of radiation to tumors with reduced exposure to normal tissues[3]. The CC49-IgG-IL-2 fusion (SCIg-IL-2) delivers IL-2 directly to the tumor microenvironment, potentially enhancing local immune responses while reducing systemic IL-2 toxicity[4]. CC49 fusion proteins are investigational and have not been approved for clinical use.
