**CC49-mal-MMAE** is a preclinical, TAG-72-targeted antibody-drug conjugate developed at City of Hope for ovarian cancer research. It consists of the murine anti-tumor-associated glycoprotein 72 antibody CC49 conjugated through reduced hinge-region disulfides to the antimicrotubule payload monomethyl auristatin E using maleimido linker chemistry. The construct had a high drug-to-antibody ratio of approximately 9.5 but showed rapid blood clearance and low OVCAR3 xenograft tumor uptake relative to the corresponding bromoacetamido-linked CC49-MMAE conjugate, and it was ineffective in the reported ovarian cancer xenograft efficacy study.
