CCI-38 is a novel small-molecule inhibitor developed to target YTHDF2 (YTH N6-methyladenosine RNA binding protein 2), a protein implicated in the progression and immune evasion of aggressive blood cancers, particularly B cell malignancies such as leukemia and lymphoma. Overexpression of YTHDF2 enables cancer cells to evade CAR T cell therapy by suppressing antigen expression (notably CD19 and HLA-DMA/B) and supporting tumor growth through enhanced energy production. By inhibiting YTHDF2, CCI-38 restores antigen expression on malignant cells, sensitizes them to CAR T cell therapy, reduces tumor burden, and may help overcome resistance mechanisms like antigen escape. The drug is being developed by City of Hope National Cancer Institute and is currently in preclinical development for hematologic neoplasms[1][2][3][4][6][8].
