Overview

The CCL22 peptide vaccine is a preclinical-stage cancer immunotherapy developed by IO Biotech. It consists of peptides derived from the signal sequence of the chemokine CCL22 (C-C Motif Chemokine Ligand 22). The vaccine is designed to activate and expand CCL22-specific effector T cells, primarily CD8+ T cells, which then target and reduce CCL22-expressing cells in the tumor microenvironment (TME). CCL22 is a key chemokine involved in the recruitment of immunosuppressive regulatory T cells (Tregs) via the CCR4 axis. By reducing CCL22 levels, the vaccine aims to decrease Treg infiltration and promote a more pro-inflammatory TME, characterized by increased infiltration of CD8+ cytotoxic T cells and M1 macrophages. Preclinical studies in mouse models of colon carcinoma, melanoma, and pancreatic cancer have demonstrated that this approach can delay tumor growth and enhance survival.
