Overview

CCL5-siRNA is a small interfering RNA (siRNA) therapeutic candidate designed to silence the expression of the chemokine CCL5 (also known as RANTES). In epithelial ovarian cancer (EOC), particularly PD-L1-negative subtypes, CCL5 is upregulated and plays a critical role in immune evasion by recruiting and activating FOXP3+ regulatory T cells (Tregs). This recruitment creates an immunosuppressive microenvironment that inhibits the activity of cytotoxic CD8+ and helper CD4+ T cells. By utilizing RNA interference to degrade CCL5 mRNA, CCL5-siRNA reduces Treg-mediated suppression and restores the production of pro-inflammatory cytokines like IFNγ, potentially enhancing the efficacy of immunotherapies in EOC. Preclinical research at the Yale University School of Medicine has demonstrated that silencing CCL5 with siRNA can effectively reverse tumor-induced immune dysfunction in co-culture models.
