CCM-EGX is a proteolysis-targeting chimera (PROTAC) being developed by Chakrabarti Advanced Technology and its subsidiary CCM 5Prime Sciences for the treatment of EGFR-mutant cancers. The therapeutic is designed to induce the selective degradation of multiple oncogenic forms of the epidermal growth factor receptor (EGFR), including common sensitizing mutations (L858R, del19), resistance mutations (T790M, C797S), exon 20 insertions, and the EGFRvIII variant, as well as overexpressed wild-type EGFR. By recruiting an E3 ubiquitin ligase to the target receptor, CCM-EGX promotes its polyubiquitination and subsequent degradation by the proteasome, offering a potential strategy to overcome resistance mechanisms associated with traditional tyrosine kinase inhibitors (TKIs). The program is being developed in conjunction with a companion diagnostic platform that utilizes bias-free Next-Generation Sequencing (NGS) and Fluorescence-Activated Droplet Sorting (FADS) PCR for precise patient identification and stratification. It is currently in the IND-enabling stage of development.
