CCN1 (Cellular Communication Network Factor 1), also known as CYR61, is a matricellular protein that plays a critical role in tissue remodeling and wound healing. In the context of pulmonary fibrosis (PF), CCN1 is highly expressed by transitional alveolar epithelial cells. Research from the University of Michigan suggests that CCN1 acts as a protective "brake" on fibrotic remodeling by inducing apoptosis in these pathologic transitional cells, likely through the activation of a p53-dependent intrinsic apoptosis cascade and caspase-3. Recombinant CCN1 is being investigated as a potential therapeutic agent to limit the aberrant accumulation of transitional cells and improve clinical outcomes in patients with lung fibrosis.
