CCN1 neutralizing antibodies are a class of therapeutic agents designed to inhibit the activity of Cellular Communication Network Factor 1 (CCN1), also known as Cysteine-rich angiogenic inducer 61 (CYR61). CCN1 is a matricellular protein that plays a critical role in cell adhesion, migration, proliferation, and the regulation of extracellular matrix production. It is frequently overexpressed in chronic inflammatory and fibrotic conditions, such as liver, kidney, and lung fibrosis, as well as in various cancers where it promotes tumor-associated angiogenesis and metastasis. By binding to and neutralizing CCN1, these antibodies aim to suppress the activation of myofibroblasts and reduce the accumulation of fibrotic tissue. Preclinical research, notably involving clones such as 093G9, has demonstrated the potential of these antibodies to mitigate organ fibrosis and inflammatory responses in animal models, although no specific candidate has yet advanced to late-stage clinical development.
