Overview

CCR4 CAR T cells are a form of chimeric antigen receptor (CAR) T-cell therapy engineered to target the C-C chemokine receptor type 4 (CCR4), which is highly expressed on malignant cells in several mature T-cell malignancies such as adult T-cell leukemia/lymphoma (ATLL) and cutaneous T-cell lymphoma (CTCL). These autologous or allogeneic cell therapies involve genetically modifying a patient's or donor's own T lymphocytes to express a synthetic receptor that recognizes and binds to CCR4 on tumor cells. Upon binding, the modified CAR-T cell becomes activated and kills the cancer cell. Notably, fratricide among these engineered cells selectively depletes Th2, Th17, and regulatory (Treg) subsets while sparing Th1 and CD8+ populations. Preclinical studies have shown robust expansion of these modified lymphocytes with potent antitumor activity against CCR4-expressing tumors[1][2]. The approach is under investigation for treating relapsed/refractory mature T-cell malignancies.
