CCR9 chimeric antigen receptor T cells are an investigational autologous cell-based immunotherapy developed for the treatment of T-cell acute lymphoblastic leukemia (T-ALL). The therapy consists of a patient's own T cells genetically engineered to express a chimeric antigen receptor (CAR) targeting the C-C chemokine receptor type 9 (CCR9). CCR9 is highly expressed in over 70% of T-ALL cases, including a high percentage of relapsed/refractory disease, while being present on less than 5% of normal T cells. This selective expression profile is designed to overcome major challenges in CAR-T therapy for T-cell malignancies, specifically avoiding CAR-T cell fratricide (self-destruction) and prolonged T-cell aplasia. Preclinical studies have demonstrated robust antileukemic activity in both in vitro and in vivo models, and the therapy has advanced to Phase 1 clinical trials.
