**CCS1477 + darolutamide** is an investigational oral combination therapy for metastatic castration-resistant prostate cancer (mCRPC). CCS1477 is a small molecule inhibitor of p300/CBP, designed to downregulate expression and function of all forms of the androgen receptor (AR), including mutated and splice variants. **Darolutamide** is a nonsteroidal androgen receptor antagonist that inhibits the binding and activity of androgens at the AR, blocking AR nuclear translocation and AR-mediated transcription. Combining CCS1477 with darolutamide aims to more effectively suppress AR pathway activity by targeting independent, complementary mechanisms. The regimen is under clinical investigation for mCRPC in patients who have failed prior treatments, with a phase I/IIa trial ongoing to determine safety, dosing, and preliminary efficacy[1][5][7].
