CCT-217 is a first-in-class dual small interfering RNA (siRNA) therapy developed by Canary Cure for the treatment of obesity. It targets two genes—cannabinoid receptor 1 (CB1R), part of the endocannabinoid system involved in energy balance and fat storage, and zinc finger protein 423 (ZFP423), which plays a role in adipocyte differentiation. By silencing these genes through RNA interference, CCT-217 induces significant weight loss, reduces visceral and subcutaneous fat depots, preserves lean body mass, improves metabolic health markers such as cholesterol and inflammation levels, and promotes browning of white adipose tissue. The therapy is designed to be peripherally restricted to avoid central nervous system side effects associated with CB1R modulation. Preclinical studies have demonstrated substantial efficacy with potential for infrequent dosing (single or twice-yearly). As of early 2025, it is in preclinical development with Phase I trials planned for obesity.
