CCT301-59 is an investigational autologous **ROR2-targeted conditionally active CAR-T cell therapy** developed using conditionally active biologic technology. Patient T cells are genetically modified with a lentiviral vector to express a CAR that recognizes receptor tyrosine kinase-like orphan receptor 2 on tumor cells. The CAR is designed for preferential activation under acidic tumor-microenvironment conditions, aiming to reduce on-target off-tumor activity while enabling CAR-T-cell-mediated killing of ROR2-positive cancer cells. It was evaluated in recurrent or refractory metastatic renal cell carcinoma and other ROR2-positive solid tumors, including bladder cancer, gastric cancer, pancreatic cancer, and soft tissue sarcoma; the registered studies were terminated.
