CCW702 is a novel bispecific antibody designed as an immunotherapeutic for metastatic castration-resistant prostate cancer (mCRPC). It consists of a small molecule ligand (DUPA) that specifically targets prostate-specific membrane antigen (PSMA), conjugated to an anti-CD3 antibody via site-specific unnatural amino acid chemistry. This structure enables the redirection of T cells to PSMA-expressing tumor cells, resulting in T cell activation and targeted cytotoxicity against prostate cancer. The drug was developed by Calibr at Scripps Research and has demonstrated potent antitumor activity in preclinical models, with good tolerability and subcutaneous bioavailability. Clinical development included a phase 1 trial evaluating safety, pharmacokinetics, pharmacodynamics, and preliminary efficacy in men with mCRPC who had progressed on prior therapies[1][3][4][6].
