CCX559 is a potent, orally administered small molecule inhibitor of programmed death-ligand 1 (PD-L1), developed for cancer immunotherapy. Unlike monoclonal antibody checkpoint inhibitors, CCX559 is designed to be taken orally and offers potential advantages such as improved tumor penetration and reduced immunogenicity. Mechanistically, it selectively inhibits PD-L1 binding to both PD-1 and CD80, thereby preventing the suppression of T cell activation by tumor cells. Preclinical studies have shown that CCX559 induces dimerization and internalization of cell-surface PD-L1, further blocking its interaction with PD-1. In animal models, oral administration led to anti-tumor activity comparable to anti-PD-L1 antibodies. Early-phase clinical trials in patients with advanced solid tumors have demonstrated pharmacodynamic effects consistent with immune checkpoint inhibition—such as increased peripheral CD4/CD8 T cell proliferation and elevated plasma levels of IFNγ and related chemokines—with an acceptable safety profile[1][2][3][4][5][6].
