Overview

CCX662 is a small molecule antagonist of the CXCR7 receptor (also known as atypical chemokine receptor 3 or ACKR3), originally developed by ChemoCentryx (now part of Amgen). It targets the CXCL12/CXCR7 signaling axis, which is implicated in tumor growth, survival, and the recruitment of bone marrow-derived cells that facilitate angiogenesis. In preclinical models of glioblastoma, CCX662 has been studied for its ability to prevent adaptive vasculomegaly—a compensatory increase in vessel size that occurs when tumors are treated with VEGF inhibitors like sunitinib. By blocking CXCR7, CCX662 may enhance the efficacy of antiangiogenic therapies and reduce tumor infiltration and resistance by disrupting the recruitment of endothelial progenitor cells.
